IGFBP3 基因内含子 2 多态性及其与猪主要生长性状的关联分析

摘 要:胰岛素样生长因子结合蛋白(Insulin- like Growth Factor Binding Proteins,IGFBPs)与动物的生长发育有着密切的联系,为研究IGFBP3的多态性与猪生长性状之间的关系,本实验采用PCR-RFLP方法对大白猪、杜洛克及长白猪IGFBP3基因内含子2的多态性进行分析,并且研究了IGFBP3不同基因型对体长、体高、瘦肉率、活体背膘、肌肉厚度、0~100 kg日增重、30~100 kg日增重及100 kg体重日龄等主要生长性状的影响。结果表明:IGFBP3基因内含子2共417 bp,在3个猪群中有1个A307G多态位点与猪的生长性状有关,并且都是AG基因型占优势,杜洛克与长白猪在该位点上偏离Hardy-Weinberg平衡(P<0.010);3个品种猪群IGFBP3基因的多态性信息含量(PIC)均表现为中度多态(0.34~0.37);IGFBP3基因的GG基因型对大白猪的体高(P=0.068)、瘦肉率(P=0.058)的影响较大,对杜洛克猪的瘦肉率(P=0.099)、肌肉厚度(P=0.079)的影响较大,对长白猪的0~100 kg日增重的影响均接近显著水平(P=0.08)。GG基因型猪具有体高、瘦肉率高和生长较快的特点,为以后猪的育种和选种提供参考。

The Polymorphism of IGFBP3 Intron 2 and Its Relationship with the Main Growth Traits of Pigs

Abstract: There is a relationship between the Insulin-like Growth Factor Binding Proteins (IGFBPs) and animals’ growth and development. This research aimed to study the polymorphism of IGFBP3 and its relationship with pig growth characteristics. The method of PCR-RFLP was used to study the intron 2 of IGFBP3 of Landrace, Duroc and Landrace pigs. The influence of IGFBP3 gene type to pig body length, body height, lean meat rate, back fat, muscle thickness, 0~100 kg gain weight, 30~100 kg and 100 kg gain weight growth traits were analyzed. The results showed that there were 417 bps for the intron 2 of IGFBP3, and the polymorphism of A307G affected the pig growth. The three pigs with IGFBP3 gene heterozygous AG genotype were dominant. Duroc and Landrace were deviated from Hardy-Weinberg one quilibrium at the site (P<0.01). The IGFBP3 gene PIC showed moderate polymorphism in the three varieties of pig (0.34~0.37). The GG genotype of IGFBP3 gene had greatly influence in the body height (P=0.068) and lean meat rate (P=0.058) in large white. It had greatly influenced in the lean meat rate (P=0.099) and muscle thickness (P=0.079) of Duroc. The effect to weight gain at 0-100 kg were close to significant level in Landrace (P=0.08). This is showed that the GG genotype pig had the characteristics of higher body height and faster growth. It could suffer a suggestion for pig genetics and breeding.
